Hot migration of a VM in Hyper-V to vCenter did get an error: "The process cannot access the file because it is being used by another process".
Please help, thank you!

Welcome to StarWnd Forum!
Make sure the VM has no snapshots or active backup jobs running.
Also, make sure the VM can be moved between the datastores.

Yes, there is no snapshots on the VM and there is also no active backup jobs running.

Do mean that the Hyper-V Host (hosting the VM and where the Starwind is running from) must have access to the datastore of the vCenter (ESXi Host)?

Try moving the VM to another storage of Hyper-V host. What's the Guest OS of the VM, by the way?

Move the VM to another storage of Hyper-V host using Starwind V2V Converter or using Hyper-V Manager?
The Guest OS of the VM ist Windows 2012 R2.

Thanks for your update.
Try P2V conversion of that VM. For Windows Server 2012 it should work.
Yes, try moving it to somewhere else in Hyper-V manager.
