StarWind Virtual SAN creates a fully fault tolerant and high performing storage. Can I connect it to a Windows server 2012 R2 file server as the file storage?

Yes. There are bunch of PDFs on how to create failover SMB3 or NFSv4 shares on top of a fault tolerant block storage we create. See:
